Amg-1 is a selective IDO1 inhibitor for cancer and neurological disorder research

**Background**

Indoleamine 2,3-dioxygenase 1 (IDO1) is a rate-limiting enzyme that catalyzes the degradation of the essential amino acid L-tryptophan into kynurenine. This metabolic pathway plays a critical role in modulating the immune response, as the depletion of tryptophan and the accumulation of kynurenines can suppress T-cell activity and promote an immunosuppressive microenvironment. Consequently, IDO1 is frequently upregulated in various tumors to evade the host’s immune surveillance, making it a prime target for immunotherapy. Beyond oncology, the kynurenine pathway is implicated in hypotension and various neurological disorders. Therefore, the development of potent and selective inhibitors is essential for therapeutic intervention. In this context, we will introduce a selective IDO1 inhibitor – Amg-1.

**Definition**

Amg-1 is a potent, reversible, and selective indoleamine 2,3-dioxygenase (IDO1) inhibitor with an IC50 value of 3.0 μM.

**In Vitro Studies**

According to the Amg-1 description, this compound exhibits high selectivity for IDO1 over other related enzymes. In terms of Amg-1 biological activity, it demonstrates >80-fold selectivity over IDO2 (IC50 > 250 μM) and >20-fold selectivity over tryptophan 2,3-dioxygenase (TDO) (IC50 > 62.5 μM). The chemical properties of the molecule are defined by the Amg-1 Formula C20H16N4O3S2, with a molecular weight of 424.50. These characteristics allow the compound to effectively target the IDO1 enzyme while minimizing off-target effects on IDO2 and TDO. Research utilizing Amg-1 in vitro has focused on the purification and kinetic characterization of human indoleamine 2,3-dioxygenases to validate its inhibitory potency. In conclusion, Amg-1 is a highly selective IDO1 inhibitor suitable for research in Amg-1 Cancer, hypotension, and neurological disorders.
