Application: Anti-phospho-CREB (pSer133) antibody produced in rabbit may be used for immunoprecipitation at a working dilution of 1:50 and for immunoblotting at a working dilution of 1:1000 using an extract from SK-N-MC cells, untreated or forskolin treated. It was used for western blot analysis of proteins extracted from vascular smooth muscle cell cultures, separated by SDS-PAGE and blotted onto nitrocellulose membranes in a study. It was used to assess CREB activation by western blotting in a study.
Biochem/physiol Actions:
CREB is a phosphorylation-dependent transcription factor that stimulates transcription upon binding to the DNA cAMP response element (CRE), a sequence present in many viral and cellular promoters. Various extracellular signals including cAMP, membrane depolarization, increased intracellular Ca2+, and growth and neurotrophic factors stimulate this protein to activate transcription. Phosphorylation of CREB takes place at Ser133 and this regulates the ability of the protein to activate transcription. Studies show that CREB is a mediator in learning and long-term memory in both flies and mice.
